Selection of Post larvae for Penaeus vannamei.

Hello everyone:
      Today we will see about the importance of post larvae in success of shrimp farming. 

Success of shrimp farming depends on various factors. The most important factor is quality of post larvae. 

Different stress test, visual test, microbiological test to be done before stocking of post larvae.

Visual test:
  1.We have to observe the movement and activity of the post larvae in a small beaker or trough.
  2. Observe for reduction in swimming activity. 
   3. Stirr the water and observe for the activity of the post larvae whether it is moving against water current or moving to the centre. 
   4. Ask to the technicians about batch and serials of test done during condition and breeding time. 
   5. Ask them about weight of brooder and it's quality. 
   6. Ask for the results of PCR tested diet ( diet given during breeding)...

After asking these queries then check for microscopic observations. 
   1. Check the presence of rostral spines in the larvae. 
   2. Check the gills whether it is developed or undeveloped or fouled by any fungus. 
   3. Look for the structure of the body, ( cramp and necrosis must be absent). 
   4. Check for muscle gut ratio of the larvae. 
   5. Look over the pleopods, uropod, telson and check the colour. 
   6. Check the colour of Hepatopancreas of the shrimp, must be in brown not in white or any other colour.

Stress test:
  1. Formalin stress test with 100-150 ppm for 1-2 hrs and check for survival. It must be more than 80℅.
Note : Avoid formalin stress test for long transported seed. 
   2. Different salinity test and check for survival. 
   3. Different temperature test. 

After these test then only take sample from the stress tested animal and check for PCR ( WSSV and EHP). 
   Check for vibrio also. 

The microbiology samples must be homogenized with more than 100 pl for better results.
